PERBANDINGAN DAN KARATERISTIK NANOPARTIKEL EKSTRAK DAUN AFRIKA (Vernonia amygdalina Del.) MENGGUNAKAN POLIMER KITOSAN DAN NATRIUM ALGINAT
Abstract
Daun Afrika yang dikenal secara ilmiah sebagai Vernonia amygdalina Del. diakui mengandung beragam senyawa bioaktif, termasuk flavonoid dan seskuiterpen lakton, yang telah menunjukkan harapan yang cukup besar dalam aplikasi prospektif mereka sebagai agen untuk terapi antikanker dan antibakteri. Penelitian ini melakukan perbandingan komprehensif dari karakteristik yang disajikan oleh nanopartikel ekstrak daun Afrika, menggunakan dua polimer yang berbeda, khususnya kitosan dan natrium alginat, dengan tujuan akhir mengidentifikasi formulasi yang paling efektif untuk nanopartikel ini. Tujuan utama dari penelitian ini adalah untuk mengevaluasi dengan cermat karakteristik kualitas nanopartikel ekstrak daun Afrika yang diformulasikan dengan polimer yang disebutkan di atas dan untuk memastikan formulasi nanopartikel mana yang menghasilkan karakteristik yang paling menguntungkan dalam hal kinerja. Sebanyak empat formulasi nanopartikel berbeda disiapkan dengan menggunakan metode spray drying, yang meliputi F1 yang terdiri dari 0,5% kitosan, F2 mengandung 0,75% kitosan, F3 diformulasikan dengan 0,25% natrium alginat, dan F4 menggunakan 0,325% natrium alginat. Evaluasi yang dilakukan sepanjang penelitian ini mencakup berbagai tes. Ekstraksi ekstrak daun Afrika dilakukan melalui proses maserasi menggunakan etanol 70%, diikuti dengan fase pengeringan, setelah itu ekstrak diformulasikan menjadi nanopartikel. Hasil penelitian menunjukkan bahwa formulasi F4 muncul sebagai kandidat unggul, menunjukkan pengukuran ukuran partikel 500,1 ± 57,13 nm, nilai PDI 0,613 indikasi homogenitas, pembacaan potensial zeta -37,16 mV menandakan stabilitas, dan efisiensi enkapsulasi yang mengesankan 91,14%. Pada akhirnya, nanopartikel ekstrak daun Afrika yang diformulasikan dengan 0,325% natrium alginat, yang ditunjuk sebagai F4, mewakili formulasi optimal ketika dinilai berdasarkan parameter kritis seperti ukuran partikel, stabilitas, dan efisiensi enkapsulasi.
African leaves, scientifically known as Vernonia amygdalina Del. are recognized as containing various bioactive compounds, including flavonoids and sesquiterpene lactones, which have shown considerable promise in their prospective applications as agents for anticancer and antibacterial therapies. This study conducted a comprehensive comparison ofthe characteristics presented by African leaf extract nanoparticles, using two different polymers, specifically chitosan and sodium alginate, with the ultimate goal of identifying the most effective formulation for these nanoparticles. The main objective of this study was to carefully evaluate the quality characteristics of African leaf extract nanoparticles formulated with the polymers mentioned above and to determine which nanoparticle formulation produced the most beneficial characteristics in terms of performance. A total of four different nanoparticle formulations were prepared using the spray drying method, including F1 consisting of 0.5% chitosan, F2 containing 0.75% chitosan, F3 formulated with 0.25% sodium alginate, and F4 using 0.325% sodium alginate. The evaluations conducted throughout this study included various tests. Extraction of the African leaf extract was performed through a maceration process using 70% ethanol, followed by a drying phase, after which the extract was formulated into nanoparticles. The results of the study indicate that formulation F4 emerged as the leading candidate, showing particle size measurements of 500.1 ± 57.13 nm, a PDI value of 0.613 indicating homogeneity, a zeta potential reading of -37.16 mV indicating stability, and an impressive encapsulation efficiency of 91.14%. Ultimately, the African leaf extract nanoparticles formulated with 0.325% sodium alginate, designated as F4, represent the optimal formulation when evaluated based on critical parameters such as particle size, stability, and encapsulation efficiency.
