KLORDIAZEPOKSID SEBAGAI OBAT GOLONGAN BENZODIAZEPIN: HUBUNGAN STRUKTUR HETEROSIKLIK DAN AKTIVITAS FARMAKOLOGIS (Kajian Senyawa Heterosiklik Diazepam Cincin Tujuh pada Klordiazepoksid dan Perannya sebagai Ansiolitik)

Abstract

Senyawa heterosiklik merupakan kelompok senyawa organik yang memiliki peran sangat penting dalam bidang kimia medis dan farmasi karena sebagian besar obat modern tersusun atas kerangka heterosiklik. Benzodiazepin dan quinoxalin termasuk golongan heterosiklik yang telah banyak dilaporkan memiliki aktivitas biologis, seperti sedatif, anxiolytic, antimikroba, antiinflamasi, dan antikanker. Penelitian ini bertujuan untuk mempelajari metode sintesis senyawa heterosiklik beranggota tujuh berbasis benzodiazepin dan quinoxalin melalui reaksi multikomponen dengan menggunakan ninhidrin sebagai pereaksi utama. Proses sintesis dilakukan melalui reaksi antara ninhidrin, siklo-1,3-diketon, dan 1,2-fenilendiamina dalam pelarut asam asetat dengan kondisi pemanasan refluks. Senyawa hasil reaksi diperoleh dalam bentuk padatan kristalin dengan rendemen yang cukup tinggi. Karakterisasi struktur senyawa dilakukan menggunakan spektroskopi FT-IR, NMR (^1H dan ^13C), spektrometri massa, serta analisis difraksi sinar-X kristal tunggal. Hasil karakterisasi menunjukkan bahwa senyawa yang terbentuk memiliki struktur cincin heterosiklik kompleks yang sesuai dengan senyawa target. Metode sintesis ini dinilai efisien, sederhana, dan berpotensi untuk dikembangkan lebih lanjut dalam penelitian senyawa bioaktif baru.
